Leicester ended Gloucester's 22-game 17-month unbeaten home record but the Cherry and Whites remained top of the Premiership despite their defeat.
Ollie Smith was put clear to score a converted try but Willie Walker's penalty cut the deficit at half-time.
Olivier Azam's try from a five-metre penalty line-out put Gloucester ahead.
Walker converted but Andy Goode tied the score and Ayoola Erinle's try sealed the win, although Gloucester's bonus point kept them top of the table.
Gloucester coach Dean Ryan:
"Some of our young guys have been playing at Moseley, so you've got to take your hat off to them.
"As a collective we did everything we wanted. The senior guys turned up and the younger guys flourished."
Leicester coach Marcelo Loffreda:
"But we have a long way to go. We were sharp when we were attacking, but tried to control the tempo and couldn't.
"We have to work on that and get a better quality of ball out."
