The two athletes tested positive for diuretics, which are used for slimming and can also cover up the use of other drugs.
International Paralympic Committee president Robert Steadward said the names and nationalities of the athletes would be released after hearings on Sunday.
IPC medical officer Michael Riding added: "The sport is powerlifting, that's all we can say at this point.
"The B test results will be here this morning. Of course it's unusual for a B test not to confirm an A test."
Mr Riding said: "In powerlifting you can alter your weight class with diuretics and very effectively too."
He admitted to being shocked at the number of positive out of competition tests, saying: "Six out of 129 competitors is pretty high by anyone's standards."
Romania's Aurel Berbec, who tested positive for the anabolic steroid Stanozolol, Iran's Ali Mahmoudikordkheili (anabolic steroid Methandienone), Bulgaria's Radko Radev (diuretic Chlorthalidone) and Marina Diakonova of Russia (diuretic Hydrochlorothiazide) have all been banned from competition for four years.
One of the athletes has already returned home and moves are afoot to expel the other three from the competitors' village.
