The Jamaican, who won the 100m in a world record 9.69 seconds, jogged next to rival Shawn Crawford in the last 60m to win his semi-final in 20.09 secs.
Britain's Christian Malcolm, 29, came fourth in the first semi-final to make his second Olympic final.
Team-mate Marlon Devonish failed to join him after finishing seventh.
Although Bolt will be odds-on to win the event, it is not as easy to determine where the other medals will go.
Netherlands Antilles' Churandy Martina looked impressive in winning the first semi in a time of 20.11 secs, holding off Zimbabwe's Brian Dzingayi and American Walter Dix, the 100m bronze medallist.
Although defending champion Crawford battled for his second place behind Bolt in the second semi-final, he will certainly be in the mix for a medal along with American compatriot Wallace Spearmon, who came through to take third.
Welshman Malcolm was delighted to make Wednesday's final - eight years after making his first Olympic 200m final in Sydney.
"I'm happy, very happy," he told BBC Sport after clocking 20.25 secs in his race.
"Bit by bit, the time is coming down. In the final, anything can happen. As long as I'm injury free and fit I can make finals."
