Wolves' 48-47 victory at Monmore Green may not provide enough of a cushion for the return meeting on Thursday.
Wolves were 26-10 ahead but blew the chance to build a bigger advantage.
The aggregate winners will guarantee their place in the top flight for 2009 with the losers facing a play-off against the Premier League Champions.
Niels Kristian Iversen top scored for Wolves with an 11+1 haul with skipper Freddie Lindgren contributing 10+1.
But Peterborough are now favourites to condemn the men from Monmore to a relegation race-off with either King's Lynn or Edinburgh when they meet at the East of England Showground on Thursday night (1930 BST).
Meanwhile, Wolves have announced that teenage star Tai Woffenden will ride for them in 2009.
Woffenden, 18, has been a Wolves asset for two years but has spent the couple of seasons in the Premier League with Rye House.
"It's always been my intention to make the step-up," said Woffenden. "And a full-time move to Wolverhampton is what I've been looking to do."
