All-rounder Darren Sammy and batsman Travis Dowlin have been added to the Stanford Superstars line-up for the series involving England in Antigua.
The duo will replace injured all-rounder Dwayne Bravo and exciting West Indies opener Xavier Marshall.
No official reason has been given for the withdrawal of Marshall, though the West Indies Cricket Board told Cricinfo he had failed a drugs test.
Tonito Willett, on a training camp with Marshall, also tested positive.
The series culminates with the $20m winner-takes-all game between the All-Stars and England on 1 November.
Four players in the squad - Lennox Cush, Chad Hampson, Lindon James and Lionel Baker - have no international experience.

Each player on the winning side will pocket £500,000 ($1m) - a total of £5.5m - with a further £1m to be divided between their coach and the rest of the squad.
The remaining £3.5m will be split between the ECB and the West Indies Cricket Board.
Stanford, who has described Test cricket as "boring", believes Twenty20 can become the biggest team sport in the world and hopes the matches against England will help popularise it in the American market.
