Murray is gearing up for the US Open which starts later this month
Andy Murray will face Frenchman Jeremy Chardy in his opening match at the Cincinnati Masters on Wednesday.
Chardy, ranked 58 in the world, beat Germany's Florian Mayer 6-2 7-6(7-4) in Tuesday's first round encounter.
Murray won his only previous match against Chardy, beating him in straight sets in Montreal last year.
The Scot comes into this week's tournament on a high after his win over Roger Federer in last weekend's Rogers Cup final in Toronto.
In Tuesday's late matches, in-form Argentine David Nalbandian beat 15th seed Ivan Ljubicic 7-5 6-0, while American John Isner defeated Lucasz Kubot 6-2 6-3.
But there were defeats for the 12th seed Mikhail Youzhny and 16th seed Gael Monfils.
Youzhny fell to Frenchman Richard Gasquet 2-6 6-4 6-1, just after Monfils was upset by Alejandro Falla of Colombia 6-3 6-4.
Former world number four James Blake also lost out with his conqueror Denis Istomin of Uzbekistan to face Roger Federer in the next round but Blake's compatriot Mardy Fish beat Gilles Simon 7-6 7-5 and set up a match with Spanish eighth seed Fernando Verdasco.
Lleyton Hewitt advanced after Taiwan's Lu Yen-Hsun retired when 6-4 4-0 down, while Wimbledon runner-up and seventh seed Tomas Berdych of the Czech Republic beat Paul-Henri Mathieu of France 6-4 6-4.
He will next face Marcos Baghdatis, a 6-2 4-6 7-6 (7-4) winner over Brazil's Thomaz Bellucci.
