Ross slotted the retaken goal after Gordon Bulloch had bundled over to score deep into injury time.
Perpignan's Nicolas Durand and Roland De Marigny exchanged tries before Jean-Philippe Grandclaude's touchdown gave the visiting team the lead again.
Nicolas Laharrague also kicked 10 points for Perpignan but Ross and Bulloch's intervention proved vital.
Leeds played some open rugby early on and deservedly went ahead through Ross' penalty in the ninth minute.
But they were behind two minutes later courtesy of an opportunist try from scrum-half Durand, who grabbed the ball from the back of a maul and sprinted down the blind side to score.
Laharrague knocked over a penalty in the 33rd minute to make it 10-3 before the hosts reduced the deficit in the 37th minute, full-back De Marigny scoring in the left corner.
Ross missed the conversion and an early second-half penalty but made amends with a drop-goal in the 45th minute to put Leeds in front.
However, in the 53rd minute, Grandclaude picked up a pass off his bootlaces and cantered over to give Perpignan the lead.
The fly-half then added a penalty from 30 yards to make it 20-11 before Ross hit back with a goal of his own in the 72nd minute.
Bulloch forced his way over following a line-out before Ross secured victory for the hosts at the second attempt, referee Nigel Owens adjudging Perpignan had been encroaching.
Leeds director of rugby Phil Davies:
"Apart from the win, we can take a lot out of the game.
"We played well in terms of creating chances but there are quite a few things we can learn from them in terms of tactics in the Heineken Cup.
"At times they were excellent. But my guys stuck at it and some of the stuff we put together was pretty good."
