Fa'amatuainu is on a season-long loan with Scarlets from Bath
Scarlets began their LV= Cup campaign with a bonus-point victory over Leeds.
Jonny Fa'amatuainu went over to cancel out Christian Lewis-Pratt's early penalty before Leeds regained the lead through Warren Fury's try.
But from there Scarlets ran in 45 unanswered points, with Fa'amatuainu, Scott Williams and Daniel Newton all crossing before the break.
Fa'amatuainu got his third in the second half, with Aaron Shingler and Williams adding further scores late on.
Scarlets coach Nigel Davies said: "I was very, very pleased overall. We probably could argue that we could have been more clinical but a lot of boys haven't played that much together.
"There was some very encouraging stuff there but also a lot to work on which is a good position to be in.
"Jonny was superb and it was great to have a run-out for us today - there was a whole host of players who showed up well."
Leeds coach Neil Back said: "We're obviously disappointed with the result in the end.
"First-half there was a lot of pressure imposed by both teams, they shortened us up at times and played with a lot of width.
"I think in the first 30 minutes of the second-half we improved in a lot of areas and kept them out but we're disappointed with the last 10 minutes particularly with our game management."
