Loughinisland led 1-3 to 0-1 at the break but a missed O'Reilly penalty gave Rostrevor hope and Colm Murney's 52nd-minute got cut the margin to two.
However, late points from O'Reilly and John McCarthy made the game safe.
Holders Mayobridge defeated with Eoghan Woods and Adrian Barry scoring the important goals to set up a quarter-final tie with Bryansford.
Bryansford, Burren and Saval claimed victories in the weekend first-round games.
Bryansford edged out Castlewellan 0-6 to 0-5, Burren hammered Atticall 2-13 to 0-5 while Saval defeated Ballyholland 0-12 to 0-9.
The quarter-final draw is: Kilcoo v Burren; Bryansford v Mayobridge; Downpatrick v Loughinisland; Saval v Liatroim/Longstone.
