Ben O'Reilly's first-half goal helped Loughinisland beat Rostrevor 1-8 to 1-5 in Monday's Powerade Down SFC clash.
Loughinisland led 1-3 to 0-1 at the break but a missed O'Reilly penalty gave Rostrevor hope and Colm Murney's 52nd-minute got cut the margin to two.
However, late points from O'Reilly and John McCarthy made the game safe.
There will be two games on Tuesday (1900 BST) with Downpatrick facing Ballymartin at Clonduff and Kilcoo taking on An Riocht at Dundrum.
Holders Mayobridge defeated Clonduff 2-11 to 0-14 with Eoghan Woods and Adrian Barry scoring the important goals to set up a quarter-final tie with Bryansford.
Bryansford, Burren and Saval claimed victories in the weekend first-round games.
Bryansford edged out Castlewellan 0-6 to 0-5, Burren hammered Atticall 2-13 to 0-5 while Saval defeated Ballyholland 0-12 to 0-9.
Liatroim and Longstone will have to meet again on Saturday evening after drawing 1-10 to 2-7 on Sunday evening.
The quarter-final draw is: An Riocht/Kilcoo v Burren; Bryansford v Mayobridge; Ballymartin/Downpatrick v Loughinisland; Saval v Liatroim/Longstone.
