South Korea's Seon Hwa Lee shot a six-under-par 66 to take a two-stroke lead over Natalie Gulbis after the second round of the Fields Open.
The 20-year-old rookie moved to 13 under for the tournament as she seeks her first LPGA title.
Fellow rookie Julieta Granada birdied five consecutive holes on the back nine to finish in third place, one stroke behind Gulbis.
Britain's Karen Stupples dropped down the leaderboard after her round of 72.
Michelle Wie, playing in her second tournament as a fully-fledged professional, shot a 70 and was one of several players tied for ninth at seven under.
"I just really couldn't get things going," said Wie.
"But I still feel good about my game. I still confident and I'll be ready for tomorrow."
Overnight leader Lorena Ochoa made a 71 to move to nine-under.
