Brian Stock's stunning free-kick earned Doncaster their first win at Leeds in the first league meeting between the sides in 52 years.
Stock struck their winner in the 21st minute, crashing a free-kick past Casper Ankergren before the Leeds goalkeeper could move.
Dons stopper Neil Sullivan denied Jermaine Beckford by saving a powerful volley from Leeds' leading scorer.
And Sullivan also did well to hold Peter Sweeney's free-kick.
