Gary Hart secured Brighton's first win since Boxing Day to ease boss Mark McGhee's troubles at the bottom.
Paul Reid escaped down the right after 80 minutes and crossed for Hart to send a thumping shot into the top corner.
Reid had opened the scoring for the hosts, tapping in after Dean Hammond's header had come back off the post.
Leeds drew level shortly before the break when Robbie Blake converted a penalty after Reid was controversially adjudged to have fouled Eddie Lewis.
Brighton boss Mark McGhee: "There was a real cheer in the dressing-room because if the season ended tomorrow we would stay up.
"I'm hoping to strengthen the squad and be able to persuade players to join us.
"But playing at the Withdean stadium is something like the third world in footballing terms and it can add to the difficulty of attracting players."
Leeds manager Kevin Blackwell: "Brighton's fans were magnificent. They were like a 12th man and they made it difficult for us.
"Brighton treated the match like a cup tie. I'm disappointed we came away with nothing but our problems were in the final third.
"We seemed to create the possession and had plenty of the ball, but I can't remember too many chances."
