Peters had made just 65 runs in his previous seven innings at Lord's
Stephen Peters and Alex Wakely both hit centuries in a third-wicket stand of 182 as Northants ended day two on 280-4 chasing Middlesex's total of 347.
Peters hit 155 not out off 231 balls with 13 fours, to add to his Championship tally of three centuries and three fifties so far this season.
Wakely's 108, including 15 fours and off 193 deliveries, was only his second hundred in first-class cricket.
But he offered his wicket up cheaply, chipping to mid-on off Rob White.
Middlesex had used the new ball to good effect when Pedro Collins took care of Ben Howgego leg before on 23.
Then new county cap Gareth Berg saw Mal Loye caught in the slips to end an agonising 22-ball duck.
But Peter and Wakely forged a solid partnership through 54 overs.
Earlier Middlesex added six runs to their overnight total of 341-9 before Sri Lanka left-armer Chaminda Vaas had Collins caught to end with impressive figures of 4-49 off just 21.2 overs.
