Glamorgan youngster Ben Wright hit a career-best 172 to help his side get on top in their derby game against rivals Gloucestershire at Sophia Gardens.
James Allenby then took over hitting 105, backed by 63 from Robert Croft in a seventh-wicket stand of 153 to enable the home wide to earn a healthy lead.
After resuming on 257-3, Glamorgan were able to declare on 583-9, 166 in front.
And they then made inroads before the close, reducing Gloucestershire to 12-1 when Dean Cosker bowled Chris Dent.
Allenby's ton was his first for Glamorgan, while Wright's was only the second of his career as he dominated a fourth-wicket partnership of 157 with Michael Powell (37).
After a 30-minute delay to the third day because of rain Wright built on his fine overnight platform of 77, reaching his second century before overtaking his previous career-best of 108 (made against Leicestershire in 2007).
But he was not content with just a century as he hit Vikram Banerjee and Gemaal Hussain for sixes into the grandstand racing to 150 with the final ball before lunch - his third 50 coming up in only 49 balls.
And he was in sight of becoming Glamorgan's first double-centurion since July 2006 when he edged a Jon Lewis lifter.
After Mark Wallace went for a duck caught at third slip off Anthony Ireland it was left to Allenby, passing 50 for the sixth time in as many Championship games this season, and Croft to take Glamorgan to maximum batting points.
They then on well past Gloucestershire, for whom Ireland was the pick of the bowlers, taking 5-114.
