Michael Powell struck his first century in the Championship for two years to put Glamorgan in a commanding position against Leicestershire.
Powell's 120 from 234 balls, allied to contributions from Mark Wallace (59) and Jason Gillespie (34) helped Glamorgan to a first-innings 450.
Leics were soon reduced to 66-4 in reply, with David Harrison wiping out their top order by taking 3-31.
Josh Cobb and Paul Nixon put on 51 to guide Leics to 117-4 at stumps.
Glamorgan resumed on 265-4 and Powell took his overnight 80 to three figures - his first since scoring 299 against Cheltenham in 2006.
The Welsh side passed 400 for the first time this season and they put Leics in all kinds of trouble when Harrison removed Matthew Boyce with his second ball.
Greg Smith (19), Boeta Dippenaar (eight) and HD Ackerman (14) also lost their wickets before the close of play.
