Lance Klusener's maiden double century put Northants in a strong position after three days at Wantage Road, as Glamorgan closed 173 behind at 28-1.
Niall O'Brien, who resumed on 99, made 168, eight short of his top first-class score, sharing 211 with Klusener, who was dropped twice on the second day.
Monty Panesar hit four fours in his 30 and Klusener reached his 200 with his 27th four before declaring 201 ahead.
He then caught Gareth Rees at mid-on for 13 before bad light ended play.
O'Brien and Klusener peppered the boundary with some superb strokeplay.
The Irish wicketkeeper took 14 from one Robert Croft over with two fours and a six.
In sight of his highest score he took a wild swipe at Dean Cosker and was stumped.
Klusener continued the barrage of runs, however, and hit Alex Wharf for four fours en route to his momentous double hundred.
