A defiant unbeaten 71 from skipper Robert Croft inspired a Glamorgan revival, with Surrey closing 209 ahead at 45-3 after two days at The Oval.
Surrey added 36 with their remaining three wickets and then reduced Glamorgan to a paltry 81-6.
But Croft shared 59 with David Harrison (40) and 70 with Dean Cosker to take his team to 269, a deficit of 164.
Surrey chose not to force the follow-on and lost three quick wickets, Scott Newman in the final over of the day.
Glamorgan lost Mark Cosgrove in the fourth over of their reply when Ian Salisbury took a slip catch at the second attempt.

leg-spinner Chris Schofield his first Championship wicket since 2004 when Richard Grant clipped to mid-wicket.
But Surrey saw Jon Batty mis-time a pull to mid-on for a duck, Mark Ramprakash beaten by a ball that kept low and Newman caught at short-leg.
