Mark Pettini and Ravi Bopara shared a third-wicket stand of 170 to put Essex 69 ahead of Surrey at 399-6 on day two.
The stand comprised 50 overs before Pettini pulled to long-leg three short of his second century of the campaign.
Bopara, who took his first five-wicket haul on day one, smashed 12 fours and a six before he was trapped lbw for 94 when attempting to hit over mid-wicket.
James Foster continued the momentum with his fifth half-century of the season, smashing 10 fours in his 71.
A difficult day for Surrey was compounded by an injury to leg-spinner Ian Salisbury.
He twisted his knee attempting a catch and spent most of the afternoon off the field, but did indicate that he would be fit to resume later in the match.
