West Indian Ramnaresh Sarwan was dismissed one run short of a second Championship ton, as Gloucestershire built a slender lead of 181 at 294-6.
Mark Ramprakash added only seven to his overnight 185 but number nine Tim Murtagh went on to 66 containing 10 fours and a six as Surrey made 463.
Sarwan hit 15 fours and a six and shared a fluent 151 with Kadeer Ali.
But four wickets for 59 put pressure on the visitors and much will depend on skipper Alex Gidman (40) on day four.
Surrey did not look likely to break the Sarwan-Kadeer partnership, but Nayan Doshi found some turn in his second over and clean bowled Ali.
Sarwan moved into the nineties with a six over extra cover, but after facing another 30 deliveries he swept straight to square leg.
