Michael DiVenuto restored some pride for Derbyshire but Gloucestershire can expect victory over the Championship's bottom side on day three.
The Tasmanian watched from the other end as fellow Aussie Ian Harvey dismissed three batsmen lbw, the second Mohammad Kaif, leaving the hosts 39-3.
But his third century of the season took the Peakites to a tenuous lead of 38 by the time bad light stopped play early.
In making an unbeaten 116 he shared stands of 106 with Dominic Hewson and an unbroken 81 with Luke Sutton, who was 38 not out at stumps.
Gloucestershire took their first innings lead to 188 on the second morning, Martyn Ball reaching 34 not out in a total of 288.
