Captain Andy Strauss led the way with a century as Middlesex enjoyed a profitable opening day at Southgate.
The opening batsman, who won the toss, stroked an impressive 147, a knock which included 21 boundaries and a six.
He shared in partnerships of 89 with Ben Hutton (40) and 92 with Ed Joyce, who was unbeaten on 81 at stumps with a century in his sights.
Strauss was eventually caught behind off left-arm spinner Rupesh Amin after a knock that spanned 290 minutes and 240 deliveries.
The Leicestershire attack, which utilised eight different bowlers, was best served by David Masters, who returned 3-71.
