Yorkshire dominated with bat and ball at the County Ground to leave the home side facing the prospect of a heavy defeat.
Craig White compiled a superb 173 not out, aided chiefly by Steve Kirby, who resisted for almost an hour and a half and contributed 33 to a last-wicket partnership of 96.
Kirby then tore into Derbyshire's top order, removing Michael Di Venuto, Chris Bassano and Mohammad Kaif.
Dominic Hewson followed Bassano and Kaif in being dismissed for a duck when he was caught by Stephen Fleming from the bowling of Ryan Sidebottom.
And the left-armer added three more wickets for figures of
4-36 before Nathan Dumelow produced a much-needed show of defiance in making an unbeaten 20 off 27 balls.
Yorkshire had earlier added 130 for their final three wickets with White collecting two sixes and 20 fours in his 398-minute innings.
Graeme Welch was the pick of the Derbyshire bowlers, returning taking of 6-102 from 41 overs.
