Lancashire stepped on the gas in the afternoon session against Leicestershire before posting a big total at Aigburth.
Resuming play on 210-1, Lancashire added 73 runs in the morning session before clubbing a staggering 220 runs in the afternoon and declaring at tea.
Stuart Law made 82 and former West Indies captain Carl Hooper a quickfire 74, while Andrew Flintoff added an explosive 71 not out off 55 balls.
Leicestershire skipper Phil DeFreitas used nine bowlers in a futile effort to turn the tide.
And DeFreitas was forced to look on as his side lost four wickets before stumps, making the follow-on mark look a long way away.
Peter Martin grabbed two wickets, including David Masters for a duck in the dying overs.
