The movie was released in the US on Wednesday, and estimates predict it's made £85.6m since then, a new five-day North American record.
Sony, who distribute the film, haven't confirmed this, but it took at least £35.m in its first two days.
The webslinging flick, starring Tobey Maguire and Kirsten Dunst, toppled the previous record holder, Shrek 2, which took £71.6m in five days.
It also shattered Return of the King's £19m record for a Wednesday opening, which was set last December.
Serious Spidey fans wore red and blue suits to some of the 4,152 cinemas it opened in on Wednesday.
But UK fans of the wall-climbing hero don't have long to wait, as Spider-Man 2 opens here on 15 July.
