The protest - organised by the Union of Country Sports Workers - briefly brought traffic to a standstill as they moved from Hyde Park towards Parliament via Whitehall.
They were greeted outside the House of Commons by pro-hunting MPs including former Labour minister Kate Hoey and Conservative MPs James Gray and Hugo Swire.
Ms Hoey said she believed many of her fellow Labour members did not see fox-hunting as a "burning issue".
"I think the prime minister realises there are a huge number of problems that this country faces," she said.
And she warned that the pro-hunting march planned for September would be huge.
Mr Gray said he welcomed the protest saying that it was important for people in favour of hunting to show that they were not prepared to give up on the issue.
"It's saying to the Labour government that this is a question of people's livelihoods."
