The bomb exploded as guests left a ceremony traditionally held at the beginning of a film-shoot. A number of prominent local politicians are among those starring or associated with the film.
Among the injured are the popular actor Mohan Babu, who is also a member of India's upper house of parliament, and the film producer Patela Ravi, a politician and former left-wing militant.
The explosion was so powerful that the car is believed to have completely disintegrated. Six vehicles were caught in the blast, including one containing a seven-member television crew, who were all killed. It also damaged several surrounding buildings and the film studio.
"The bomb was set off by remote control," said Madhava Reddy, Interior Minister of Andhra Pradesh state, of which Hyberabad is the capital.
Nobody has claimed responsibility for the bomb, but police said they suspected political rivals of Patela Ravi.
They also have not ruled out the involvement of left-wing Naxalite rebels.
Andhra Pradesh state has been the victim of a violent campaign by outlawed Maoist insurgents.
Seven policemen died and six were wounded on Monday in a landmine explosion about 220 km (140 miles) north of Hyderabad in the fourth such attack in the past two years.
The Maoist People's War Group (PWG) was banned in Andhra Pradesh in 1992 but gained a short reprieve in June 1995 when local authorities granted a temporary amnesty to its members. The group was banned again in July last year.
