Among the highlights of the Inside Out event will be an outdoor performance by the Philharmonia Orchestra.
There will also be screenings by the National Film Theatre, performances by the National Theatre and skateboarding demonstrations all in the open air.
Mayor Ken Livingstone said the weekend was a chance to show Londoners what the area south of the Thames has to offer.
As well as music, food, books and stalls from Borough Market, visitors can pick up a South Bank loyalty card offering discounts on attractions, including the London Eye.
James Bidwell, chief executive of tourism organisation Visit London, said: "Over the past few years the South Bank has transformed itself into a world leading visitor destination, attracting tourists and Londoners."
