The singer, whose band's hits include The Power of Love and Stuck with You, will play lawyer Billy Flynn in the New York production from 1 November.
Best known for his music, Lewis has moved into acting in recent years.
The 55-year-old was Gwyneth Paltrow's father in Duets and had a supporting role in Robert Altman's Short Cuts.
Lewis will join the Broadway production just as Brooke Shields leaves it.
The actress, currently playing Roxie Hart in the London staging, will star in the New York version from 9 September to 30 October.
Lewis will play the role Richard Gere played in the 2002 film version. Gere went on to win a Golden Globe for his performance.
