Michelle Rodriguez - Ana Lucia in the series - and Cynthia Watros, who plays Libby, were stopped on the same road in Hawaii within 15 minutes of each other.
They were both charged with driving under the influence of alcohol and released on $500 (£288) bail each. They must each attend court on 29 December.
Both women joined the second series of the drama, which is filmed in Hawaii.
The actresses were stopped in separate cars in the early hours of Friday after their vehicles were spotted by police weaving on a road in Kailua.
They were both breathalysed and then arrested.
